This fifth part of the polyploidisation program reports the experiments that were conducted in 1993 and that were devoted to : - monitoring growth performance of Ostrea edulis diploids and triploids in La Trinité sur Mer and Palavas - monitoring growth performance of Crassostrea gigas diploids and triploids in Port en Bessin, Marennes Oleron (2 locations), Arcachon and Palavas - comparative study of gametogenesis for C. gigas diploids and triploids - comparative study of C. gigas diploids and triploids using sensorial analysis At the end of fifth year, it was clear that triploids are advantageous although such a statement must be moderated according to locations. The gametogenesis stydy showed that triploidy is associated with partial to complete... |

Genetic variability at five microsatellite loci was analysed in three hatchery-propagated populations of the flat oyster, Ostrea edulis. These populations were part of a selection programme for resistance to the protozoan parasite Bonamia ostreae and were produced by mass spawns, without control of the genealogy. Evidence for population bottlenecks and inbreeding was sought. A reduction in the number of alleles, mainly due to the loss of rare alleles, was observed in all selected populations, relative to the natural population from which they were derived. Heterozygote excesses were observed in two populations, and were attributed to substructuring of the population into a small number of families. Pedigree reconstruction showed that these two populations... |

Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Results were congruent between allozymes, microsatellites and mitochondrial DNA and support an isolation by distance model. High variability was observed in the breadth of diversity between populations using a mitochondrial marker, this highlights the small effective population sizes in some locations. A 10-fold quantitative difference was observed in Fst... |

Some observations on spat collection, carried out in 1976 and 1977, lead us to use mussel shells in suspension. In 1978, while bottom collection, in deep water farms, had been very bad, large quantities of spat were collected using our method. The sectors used, the percentage of collecting mussel shells and the amount of spat par shell are indicated in this note. |

A breeding programme has been undertaken by Ifremer to produce selected Ostrea edulis families which are resistant to Bonamia ostreae, a parasite which is the most significant constraint on aquaculture of this oyster. Using such families, we aimed to identify Quantitative Trait Loci (QTLs) of resistance/susceptibility to bonamiosis. We used an F2 family derived from a bi-parental cross between a wild oyster and an individual from one of the selected oyster families. A total of 550 F2 individuals from the family cohabited with wild oysters that were injected with the parasite. Mortality was checked daily over six months and heart smears were used on moribund or dying oysters to detect the presence and level of infection of the parasite. Ninety two F2... |
